Which of the following DNA sequencing method uses luciferase to generate light for detection of the individual nucleotide added to the nascent DNA, and the combined data are used to generate DNA sequence ?
Answer: C
The method is Pyrosequencing.
In pyrosequencing, nucleotides are added one type at a time. Each time one is incorporated, pyrophosphate is released; ATP sulphurylase turns it into ATP, and luciferase uses that ATP to emit light. The flash shows which nucleotide was added, and the series of flashes gives the sequence.
The Maxam-Gilbert method (spelled Maxum in the paper) uses base-specific chemical cleavage, chain termination (Sanger) uses dideoxynucleotides read on a gel, and dye-terminator sequencing labels those dideoxynucleotides with fluorescent dyes. None of them uses luciferase.
