What is the drawback of derivatisation?
Answer: B
Any incomplete or variable reaction introduces an error that the chromatography cannot detect, so the derivatisation must be reproducible to the same standard as everything else. The analyte is modified rather than destroyed, mass spectrometry works well on derivatives, and the phase is chosen freely. Adding a chemical step to a physical method imports the chemistry's variability, which is why derivatisation is avoided where an alternative exists.
